چکیده

MOTIVATION Fluorescent and luminescent reporter gene systems in combination with automated microplate readers allow real-time monitoring of gene expression on the population level at high precision and sampling density. This generates large amounts of data for the analysis of which computer tools are missing to date. RESULTS We have developed WellReader, a MATLAB program for the analysis of fluorescent and luminescent reporter gene data. WellReader allows the user to load the output files of microplate readers, remove outliers, correct for background effects and smooth and fit the data. Moreover, it computes biologically relevant quantities from the measured signals, notably promoter activities and protein concentrations, and compares the resulting expression profiles of different genes under different conditions. AVAILABILITY WellReader is available under a LGPL licence at http://prabi1.inrialpes.fr/trac/wellreader.

Investigation of an Optimized Context for the Expression of GFP as a Reporter Gene in Chlamydomonas Reinhardtii

Background: Chlamydomonas reinhardtii is a novel recombinant eukaryotic expression system with many advantages including fast growth rate, rapid scalability, absence of human pathogens and the ability to fold and assemble complex proteins accurately, however, obstacle relatively low expression level necessitates optimizing foreign gene expression in this system. The Green Fluorescent Protein (G...

Human papillomavirus genotype 16 pseudovirus production and purification in HEK-293FT cells

Introduction: Human papillomavirus (HPV) is the main causative agent of cervical cancer worldwide leading to a big health problem, especially in the developing countries. Among 14 common high-risk genotypes, HPV16 accounts for more than 50% of all cervical cancers. The current prophylactic vaccines against HPV infection are based on L1 protein. Due to some drawbacks in the current vaccines such...

A New Reporter Gene Technology: Opportunities and Perspectives

The paper summarizes the current status of the reporter gene technology and their basics. Reporter gene technology is widely used to monitor cellular events associated with gene expression and signal transduction. Based upon the splicing of transcriptional control elements to a variety of reporter genes, it “reports” the effects of a cascade of signaling events on gene expression inside cells. ...

USE OF ORGANIC SOLVENTS FOR THE ENHANCEMENT AND LIFETIME INCREASING OF LUMINESCENCE IN FLUORIMETRIC APPLICATION BY FLUORESCAMINE

In this study, the fluorogenic efficiency of fluorescamine for spectrofluorimetric analysis of primary amines was investigated. Different mixtures of dichloromethane and other organic solvents were used for evaluation of fluorescence stability and intensity of fluorescamine derivative (Pyrrolinone). Solvents such as acetonitrile (ACN), tetrahydrofuran (THF) and dioxane increase stability an...

مهار بیان ژن GFP به وسیله تداخل RNA (RNAi) در دودمان سلولی کارسینومای جنینی P19

Introduction: RNA interference (RNAi) is a phenomenon of gene silencing that uses double-stranded RNA (dsRNA), specifically inhibits gene expression by degrading mRNA efficiently. The mediators of degradation are 21- to 23-nt small interfering RNAs (siRNA). The use of siRNAs as inhibitors of gene expression has been shown to be an effective way of studying gene function in mammalian cells.  Ai...
